Dophilin (道福寧) 高血壓藥
成份 ：Doxazosin mesylate
適應症：良性前列腺肥大、高血壓。
[用法/用量]
高血壓：每日1-16mg，起始劑量為1mg，一天一次，然後按照病人的反應調整劑量，一般使用的劑量為每天2-4mg。
 良性前列腺肥大： 每日1-8mg，起始劑量為1mg，一天一次，然後按照病人的反應調整劑量。在給藥期間，必須定時評估病人的血壓。
適應症 ：Hypertension; benign prostatic hyperplasia (BPH).
用量 ：Hypertension Initially 1 mg daily, may be increased after 1-2 weeks to 2 mg once daily, & thereafter to 4 mg once daily. Max: 16 mg daily.  BPH Initially 1 mg daily, dose may be doubled at intervals of 1-2 weeks, up to a max of 8 mg daily.
用法 ：May be taken with or without food

Category C: Either studies in animals have revealed adverse effects on the foetus (teratogenic or embryocidal or other) and there are no controlled studies in women or studies in women and animals are not available. Drugs should be given only if the potential benefit justifies the potential risk to the foetus.

Renal or hepatic impairment, heart failure, pregnancy, lactation.

INDICATIONS
       Cardura is used for treating high blood pressure and treating signs and symptoms of benign prostatic hyperplasia (BPH). Cardura is an alpha-blocker. It works by relaxing muscles in the blood vessels, resulting in lowering of blood pressure. In men with BPH, alpha-blockers work by relaxing muscles around the urethra (the tube that drains urine from the bladder), which improves urinary symptoms.
INSTRUCTIONS
       Use Cardura as directed by your doctor.
• Take Cardura by mouth with or without food.
• Cardura may cause a sudden drop in blood pressure after the first dose. Take your first dose at bedtime. If you get up during the night, sit up slowly, then stand slowly.
• If you miss a dose of Cardura, take it as soon as possible. If it is almost time for your next dose, skip the missed dose and go back to your regular dosing schedule. Do not take 2 doses at once.
       Ask your health care provider any questions you may have about how to use Cardura.
STORAGE
       Store Cardura at 77 degrees F (25 degrees C). Brief storage at temperatures between 59 and 86 degrees F (15 and 30 degrees C) is permitted. Store away from heat, moisture, and light. Do not store in the bathroom. Keep Cardura out of reach of children and away from pets.

Doxazosin mesylate
       Doxazosin mesylate, a quinazoline compound sold by Pfizer under the brand names Cardura and Carduran, is an α1-selective alpha blocker used to treat high blood pressure and urinary retention associated with benign prostatic hyperplasia (BPH).
       On February 22, 2005, the US FDA approved a sustained release form of doxazosin, to be marketed as Cardura XL.
       It is an alpha-1 adrenergic receptor blocker that inhibits the binding of norepinephrine (released from sympathetic nerve terminals) to the alpha-1 receptors on the membrane of vascular smooth muscle cells. The primary effect of this inhibition is relaxed vascular smooth muscle tone (vasodilation), which decreases peripheral vascular resistance, leading to decreased blood pressure.
       Doxazosin and similar medications like prazosin have been found to help reduce the intensity of and/or stop posttraumatic stress disorder night terrors and nightmares. The full reasoning of this effect is not understood.
 Efficacy
      In March 2000, the Antihypertensive and Lipid Lowering Treatment to Prevent Heart Attack Trial (ALLHAT) study stopped its arm of the trial looking at alpha blockers, because doxazosin was less effective than a simple diuretic, and because patients on doxazosin had a 25% higher rate of cardiovascular disease and twice the rate of congestive heart failure as patients on diuretics. Pfizer, aware of the results before publication, launched a marketing campaign in early 2000, and sales were largely unaffected, despite the dangers highlighted by the study.
       Doxazosin shows potential for treatment of benign prostatic hyperplasia and erectile dysfunction.
      Doxazosin has also shown some efficacy in treating chronic epididymitis.
